Berthon began his international karting career in 2006, finishing third in the Copa Campeones Trophy ICA class and fifth in the French Championship Elite category. He repeated the fifth-place result in the French Elite class the following year and also competed at the South Garda Winter Cup and the KF1 World Cup.
In 2008, Berthon moved into single-seaters with Boutsen Energy Racing, contesting both the Formula Renault 2.0 West European Cup and the Eurocup Formula Renault 2.0. He scored points on four occasions in the West European Cup and finished 18th overall, while failing to score in the Eurocup.
He remained in both series for 2009 with Epsilon Euskadi. In the Eurocup, he took a victory at the opening round in Barcelona after teammate Albert Costa was disqualified for a technical infringement, then added a further podium at the Hungaroring to finish sixth in the championship. In the West European Cup, Berthon won at Spa-Francorchamps and took six additional podium finishes to finish third in the standings behind Jean-Éric Vergne and champion Costa. He was also crowned French Formula Renault 2.0 champion that season, the series having run as part of the West European Cup calendar.
Berthon tested a Formula Renault 3.5 car in October 2009 for Tech 1 Racing at Motorland Aragón as a reward for his French Renault title. He subsequently drove for the recently crowned champions Draco Racing at the Circuit de Catalunya test and was announced as their driver for the 2010 season, partnered by Colombian driver Omar Leal.
In 2010 with Draco Racing, Berthon finished on the podium at the season-opener at Motorland Aragón and at Brno before taking his first Formula Renault 3.5 victory at Magny-Cours, holding off a strong challenge from Daniel Ricciardo. A further podium at Hockenheim saw him classified seventh in the championship. He returned to the series in 2011 with ISR Racing, replacing Dean Stoneman after the Briton was forced out through illness, and finished 13th overall.
In July 2010, Berthon made his Formula Three debut with ART Grand Prix in the British Formula 3 Championship round at Spa-Francorchamps, entered in the Invitation class. His best result over the race weekend was 13th place.
Berthon debuted in the GP2 Asia Series in 2011, partnering Dani Clos at Racing Engineering, but did not score a point in the four races he contested. He returned with the same team for the non-championship race concluding the 2011 season and then raced full-time alongside Fabio Leimer in the 2012 GP2 season. He finished 12th in the 2012 standings with two second-place finishes as his best results. During the winter of 2012 he also competed in the Toyota Racing Series in New Zealand with M2 Competition, finishing seventh in the championship.
On 16 November 2011, Berthon drove a Formula One car for the first time, taking the wheel of the HRT F111 at the Abu Dhabi young drivers' test at Yas Marina Circuit. He completed nine laps on the first day, posting a best time of 1:48.646 to rank 13th fastest. The following day he had sole use of the car, completing 51 laps over 283 kilometres and improving his best time to 1:45.839 to finish 12th.
In October 2015, Berthon was announced as a race driver for Team Aguri for the 2015-16 Formula E season, partnering António Félix da Costa. He was replaced by Salvador Durán after three races and has not competed in Formula E since.
Berthon has gone on to compete in endurance racing, including the 24 Hours of Le Mans and the FIA World Endurance Championship, as well as touring car categories including the World Touring Car Cup and TCR Europe Touring Car Series.
